Bruce
Merrifield's original synthesis of peptides attached protected amino
acids to a growing chain that was covalently bonded to a resin. That
unheard of method to synthesize peptides not only won him the Nobel
Prize in Chemistry in 1984,
but also ushered in a new era of peptide research - scientists were now
able to design their own peptides and study them at a previously
unattainable scale. Nowadays, you can still do it yourself (see our
section on peptide
synthesis reagents)
or you can order them from a variety of companies as single purified
entities, in the form of libraries, optimized for anti-peptide antibody
production (MAPS), or even recombinantly produced.

custom - small scale
(typically < 1 gram) synthesis of peptides containing the
standard 20 amino acids

modified
- small scale (typically < 1 gram) synthesis of modified
peptides
(e.g. N-acetylated, C-amidated, biotinylated, etc.) or peptides
containing unusual or unnatural amino acids (e.g., phosphotyrosine,
D-amino acids, beta-amino acids, etc...). Please note that each company
has its own list of modifications it can incorporate. As a result, you
will have to check with the individual company to see what they can do.

- able to produce peptide libraries or peptide arrays for screening
purposes - check with the company for the format and degree of purity
- MAPs - able to produce
multiple antigenic peptides (MAPs)

Peptide Fun Fact:
The Fmoc group is the most common amino acid protecting group using
today in the solid phase synthesis of peptides. However, the
use
of this group is relatively new in the history of peptide
synthesis. Peptides were originally produced more laboriously
in
solution where a strategy of amine protection followed by coupling then
amine deprotection had to be followed. Developed by Bergman
and
Zervas in 1932 [Ber. Dtsch. Chem. Ges. 65 (1932)
1192], the
Cbz group was the first amine protecting group to be used and was
removed using catalytic hydrogenolysis. The acid labile Boc
group
was developed by Carpino in 1957 [J.
Am. Chem. Soc. 79 (1957) 98]
and was the second popular protection strategy to produce peptides in
solution. The use of the fluorenylmethoxycarbonyl (Fmoc)
group,
also invented by Carpino [J.
Am. Chem. Soc. 92 (1970) 5748], became popular in
the 1980s because of its use on automated peptide synthesizers.
